Cytokine measurements in marmoset with ELISA and ELISPOT

Marmoset ELISAs

U-CyTech biosciences has developed various sensitive and specific sandwich ELISA kits and T cell ELISPOT kits for the measurement of cytokine production in the common marmoset (Callithrix jacchus).

The ELISA set-up and performance is fast and simple and suitable to quantify your marmoset cytokine of interest in cell culture supernatant, serum and/or plasma. Whereas the T cell ELISPOT assay detects cytokine secreting T cells (originate from e.g. PBMC) at the single cell level in response to an antigen.

Our company has developed and validated ELISA tests for several important marmoset cytokines, such as IFN-γ, TNF-α, IL-2, IL-6, IL-12/23p40, IL-13 and IL-17A. For the T cell ELISPOT assay IFN-γ, TNF-α, IL-12/23p40, IL-13 and IL-17A are available.

 
 
The common marmoset is a small New World monkey, which is increasingly used as a preclinical animal model for human disease. This animal has proven to be a very important model for the evaluation of new drugs and therapies in man. Because of species-specificity, most drugs and therapies cannot be studied in lower animals such as mice and rats, but can effectively be studied in this non-human primate. Because marmosets are not aggressive in nature and much smaller than macaques, the animals have major benefits over the use of Old World monkeys including housing and the management of experimental condions. However, limiting factors of this animal model are the small quantities of blood and tissue available for immunoassays and lack of reagents to measure cellular and humoral immune responses.

 

Our Marmoset ELISA kits contain 96-wells plates. However for smaller size samples it is also possible to use 384-wells plate which require only 20 µl of diluted sample per well. We can recommend the 384-well plates from Greiner bio-one (cat. no. 781061).
When using a 384-well plate, we recommend to use a volume of 20 µl per well for the coating antibody solution, standard, blank, diluted samples, detection antibody solution, SPP conjugate, TMB substrate solution and stop solution. For blocking, we recommend a volume of 50 µl per well. Please check whether your plate reader is able to read such plates.
